Abstract

Le procédé de circularisation d'un oligonucléotide selon l'invention comprend: l'incubation d'un acide nucléique double brin comprenant une séquence cible, avec un oligonucléotide simple brin comprenant une partie centrale capable de se fixer sur ladite séquence cible, reliée par des séquences espaceurs à des séquences terminales en 5' et 3', ces séquences étant: soit a) complémentaires de la séquence d'une matrice oligonucléotidique; soit x) partiellement complémentaires l'une de l'autre, de manière à pouvoir s'hybrider l'une à l'autre en formant un double brin tout en conservant une extrémité en simple brin, laquelle est capable de s'hybrider à la séquence terminale d'un autre oligonucléotide, dit oligonucléotide-boucle, qui possède une structure en épingle à cheveux, avec une partie en simple brin et une partie repliée qui forme un double brin, la mise en contact, dans le premier cas a), du complexe formé avec ladite matrice oligonucléotidique, dans des conditions permettant l'hybridation des extrémités 5' et 3', de manière adjacente, à la matrice, et dans le cas x), celle du complexe formé avec ledit oligonucléotide-boucle, dans des conditions permettant, par l'hybridation des extrémités libres de l'oligonucléotide simple brin et de l'oligonucléotide-boucle, la juxtaposition des extrémités 5' et 3' de ces oligonucléotides, la ligation des extrémités hybridées, de manière à circulariser l'oligonucléotide autour de l'acide nucléique double brin et, si on le souhaite, l'isolement de la structure circularisée. Application du procédé et des structures obtenues en diagnostic et thérapeutique.
