Abstract

La présente invention se rapporte à un système de puces à ADN pour détecter une mutation dans une acide nucléique cible de maniéreà ce que seul l'ADN comportant la mutation demeure sur la puce à l'issu du procédé. L'invention concerne un procédé dans lequel un αS-phosphothioatedésoxynucléotide complémentaire de la mutation est ajouté au moyen d'une ADN polymérase à l'extrémité 3' de la sonde hybridée à l'acide nucléique cible et dans lequel une exonucléase est ajoutée de sorte que seules les sondes élongées ne sont pas dégradées. La détection de la présence ou de l'absence de la mutation est effectuée par mesure directe ou indirecte de la présence ou de l'absence d'ADN en un site donné sur la puce. Avantageusement, la puce comprend des transistors du type ISFET ou des transducteurs piézo-électriques.
