Definitions
- the present invention belongs to the field of pharmaceutical synthesis, and in particular, to a JAK enzyme inhibitor, a preparation method thereof and use thereof.
- Protein kinases also known as Protein Phosphokinase, are a class of enzymes that catalyze the phosphorylation of proteins and are key factors in regulating cell signaling, including cell proliferation and cell differentiation.
- JAK1 also known as Janus kinase-1
- JAK2 also known as Janus kinase-2
- JAK3 also known as JAKL or L-JAK, leukocyte Janus kinase; and Janus
- TYK2 also known as protein-tyrosine kinase 2
- Blocking signal transduction at the level of JAK kinase provides promise for the development of therapeutic approaches for inflammatory diseases, autoimmune diseases, myeloproliferative diseases, and cancer. Inhibition of JAK kinase also contributes to the treatment of skin immune diseases such as psoriasis and skin sensitization.
- Pfizer's Toficitinib has been marketed for the treatment of rheumatoid arthritis; and Incyte's rosotinib is used to treat myelofibrosis and acute graft-versus-host disease.
- JAK enzyme inhibitors also have some obvious side effects.
- some JAK inhibitors are prone to the following side effects: infection, including pneumonia, viral infection (such as herpes zoster infection), bacterial infection, actinomycetes Infection (mycobacterial infection), fungal infection, decreased immunity (such as decreased NK cells), and anemia.
- infection including pneumonia, viral infection (such as herpes zoster infection), bacterial infection, actinomycetes Infection (mycobacterial infection), fungal infection, decreased immunity (such as decreased NK cells), and anemia.
- Black Box is even caused by some serious side effects, such as acute tuberculosis, invasive fungal infections, bacterial infections, and some lymphomas or other tumors.
- Studies have shown that currently available JAK inhibitors tend to have inhibitory activity against both JAK1 and JAK3, and most of these side effects are associated with inhibition of JAK3 activity.
- JAKs family of kinases are responsible for the regulation of numerous signaling pathways. Since JAK1 and JAK3 are part of the common gamma-chain cytokine receptor complex, this has led to significant difficulties in developing inhibitors with high selectivity for JAK1.
- a first aspect of the invention provides a compound of formula I, or a stereoisomer thereof, or a tautomer thereof, or a pharmaceutically acceptable salt, hydrate or solvate thereof:
- X is N or O
- Y is N or O
- Z is selected from the group consisting of substituted or unsubstituted C 1 -C 8 alkyl
- W is N or C
- R 1 , R 2 , R 3 are each independently selected from the group consisting of hydrogen, substituted or unsubstituted C 1 -C 6 alkyl, substituted or unsubstituted C 2 -C 6 alkenyl, substituted or unsubstituted C 2- C6 alkynyl, substituted or unsubstituted C 3 -C 8 cycloalkyl, substituted or unsubstituted C 6 -C 10 aryl (preferably phenyl), substituted or unsubstituted having 1- 3 5-10 membered heteroaryl (preferably 5- or 6-membered), substituted or unsubstituted benzo 5-10 membered heteroaryl selected from the group consisting of the following heteroatoms of N, O, S (more Good land is ⁇ );
- R 4 is selected from the group consisting of substituted or unsubstituted C 1 -C 6 alkyl, substituted or unsubstituted C 3 -C 8 cycloalkyl, substituted or unsubstituted, having 1-3 selected from the group N, a 3-10 membered heterocycloalkyl of a hetero atom of S, O, CH 2 OR 5 , wherein R 5 is selected from the group consisting of C 1 -C 6 alkylcarbonyl, trialkylsilyl;
- substitution means substitution by one or more (eg, 2, 3, 4, etc.) substituents selected from the group consisting of: halogen (preferably F), CN, C 1 -C 6 alkyl, halogenated C 1 -C 6 alkyl, or C 1 -C 6 alkoxy.
