- Example 5 The same procedure used in Example 5 was used to transfect the 5 miRNAs found identified in Example 5. However, 17-AAG was added in a 1 :800, 1 :3200 at increasing concentration of transfected miRNAs o further define its optimal concentration for apoptotic activity. The cells were incubated for 48 hours, and developed using the same procedure as described in Example 5 and using SEQ ID NOs: 1-7. Results are shown in Figure 6.
- This Example demonstrates that 17-AAG sensitizing activity can be increased with combination of various miRNAs, specifically, miR-634 and miR-648 (using SEQ ID NOs: 1 -2).
- MiR-634 and miR-648 were tested alone and in combination for the enhancement of apoptotic activitiu using the apoptotic assay system of Example 1. Results are shown in Figure 7. Increased apopotic activity was shown for the combination of miR-634 and miR- 648 as compared to either alone.
- HT1080 cells were trypsinized from a confluent 25 cm or 75 cm flask and resuspended in 10 mL complete medium, then diluted to 25,000 cells/mL in at least 12 mL total medium volume. One hundred of cell suspension was added to each well of the 96- well ImageLock plate containing transfection reagents. Cells were allowed to settle to the bottom of the plate for 10 to 15 minutes, then were incubated at 37 °C, 6% C0 2 for 48 hours.
- IncuCyte phase-contrast compact microscope (Essen Bioscience, Ann Arbor, MI) inside a tissue culture incubator and allowed to equilibrate for 5 minutes before starting the first scan.
- the plate was scanned every 1 to 2 hours at one image per well at least until wounds in control wells were completely closed.
- InM miR-634 combined with 75 nM 17AAG showed inhibition of migration on HT1080 cells.

Abstract
Cette invention concerne des procédés et des compositions de criblage de micro-ARN pouvant moduler l'expression génique dans la voie apoptotique en la présence d'inhibiteur de HSP90. L'invention concerne également l'utilisation de miARN pour stimuler l'activité d'agents thérapeutiques non limités à un inhibiteur de HSP90, ainsi que l'utilisation en diagnostic de miARN pour prédire la réponse à une thérapie non limitée aux agents thérapeutiques. L'invention concerne finalement un procédé pour l'identification et l'application thérapeutique de petites molécules qui sont des modulateurs de ces acides nucléiques.
