- the particle size distribution was evaluated by laser diffraction analysis (Sympatec GmbH, Germany) equipped with Windox 4 software.
- Formulations Fl-6 did not show any precipitation of the active immediately after filling; however, all investigated compositions finally showed some kind of precipitate after storage for a few days or after mechanical stress (shaking).
- the most promising formulation was found to be F6 containing 17.5% ethanol, 12.5% propylene glycol and 67.5% HFA 227. This formulation did not show precipitation until day 21 after manufacture. However, after 3 weeks storage a slight precipitate was observed which was easily redispersable.
- Table 2 Formulations showing least precipitation and no / minimal glass container adsorption
- formulation F6 was freshly prepared and sprayed for aerosol droplet size measurement using different actuator orifice diameters.
- formulation F6 The droplet size distributions of formulation F6 were determined by laser diffraction (HELOS-laser diffractometer, SympaTEC GmbH, Germany). The MDI was positioned in front of the laser beam at a distance of approx. 12 cm. The measurements were performed when the optical concentration of aerosol was > 0.1. The R3 lens (measuring range from 0.5 ⁇ m to 175 ⁇ m) was used. The raw data for the determination of particle size distribution was evaluated by the Sympatec Windox 5 Software according to the Fraunhofer theory. Formulation F6 with 12.5% propylene glycol and 17.5% ethanol showed average particles sizes of roughly 25 ⁇ m and and a d90% of roughly 40 ⁇ m.

Abstract
La présente invention concerne des compositions pharmaceutiques non aqueuses à longue durée de conservation ainsi que leur utilisation dans des méthodes de traitement du diabète et de l'hyperglycémie. Ces compositions comprennent un peptide insulinotropique et un solvant organique protique semi-polaire.
