Abstract

L'invention concerne des procédés catalysés enzymatiques et des produits ainsi que des kits associés en vue d'une liaison efficace d'une molécule avec une protéine ou un peptide et les conjugués générés. Une enzyme utilisée fréquemment présente une activité transamidase, notamment une sortase ou une enzyme associée. L'invention concerne également des produits, des procédés et des kits de purification en une seule étape d'une protéine cible recombinante ou d'un peptide cible exprimé(e) à l'origine comme produit de fusion. Ce produit comporte une région d'association en phase solide, une peptidase, une séquence de reconnaissance de peptidase et une protéine cible ou un peptide cible. La peptidase peut cliver la protéine de fusion lorsque la fusion est liée à un support solide liant de manière spécifique la région d'association de phase solide, la protéine cible ou le peptide cible étant libéré(e) depuis le support solide sous forme sensiblement purifiée. La peptidase fait parfois preuve d'une activité hydrolase, notamment une sortase ou une enzyme associée par exemple.
