Abstract

Es wird ein Verfahren zur Detektion spezifischer Dinukleotide in einer DNA-Probe beschrieben. Eine Polymerase-Kettenreaktion (PCR) wird unter Verwendung a) eines Nukleotids, das Teil des zu detektierenden Dinukleotids ist, von dem eine ausreichende Menge durch ein Donor-Fluorphor markiert und b) ein anderes Nukleotid, das Teil des zu detektierenden Dinukleotids ist, von dem eine ausreichende Menge mit einem Akzeptor-Fluorophor markiert ist, durchgeführt. Das Verfahren bestimmt oder quantifiziert die Anwesenheit des Dinukleotids durch Messung des Ausmaßes des Fluoreszenzresonanzenergietransfers (FRET) zwischen dem Donor- und Akzeptor-Fluorophor.
