Abstract

Active agent-containing spherical pellets were successfully produced via a hot-melt extrusion/spheronization technique. The beads processed by this new technique were compared to beads produced by traditional wet-mass granulation/spheronization. The hot-melt extruded spherical pellets were manufactured by first cutting a thin, extruded rod into nearly symmetrical pellets. These pellets were then spheronized in a traditional spheronizer at an elevated temperature. Although the formulations were identical for beads processed by both techniques, beads displayed significantly different release patterns. The wet-mass granulated beads released active agent at a faster rate than the hotmelt extruded beads. Regardless of the solubility of the thermoformable composition comprising the beads, the hot-melt extruded beads exhibited greater control of active agent release than the wet-mass granulated beads. The difference in bead matrices is illustrated by scanning electron microscopy. Additionally, the beads processed by this new technique have a narrower particle size distribution. The new technique of hot-melt extrusion/spheronization can produce spherical particles without the addition of water or other solvents.
