Abstract

L'invention concerne une méthode permettant de détecter par électrophorèse capillaire la présence de mutations dans des échantillons d'ADN. Cette méthode consiste: (a) à introduire séparément chaque échantillon d'ADN, qui contiennent tous une solution tampon comprenant une matrice et dont au moins un contient un ADN hétéroduplex, dans une pluralité de récipients dans un système d'électrophorèse ; (b) à chauffer les échantillons d'ADN indépendamment du champ électrique dans le système d'électrophorèse dans au moins une gamme spatiale ou temporelle prédéterminée de température choisie pour dénaturer l'échantillon d'ADN ; et (c) à détecter la présence de mutations dans les échantillons d'ADN en comparant un motif électrophorétique pour chaque échantillon d'ADN avec un motif électrophorétique pour un échantillon d'ADN témoin, ce qui permet de détecter par électrophorèse multiplex la présence de mutations dans des échantillons d'ADN. Ce procédé peut également consister au point (b) à introduire séparément au moins un échantillon d'ADN témoin, qui contient un ADN hétéroduplex, dans une solution tampon comprenant une matrice, dans un récipient de la pluralité de récipients du système d'électrophorèse.
