Abstract

Pour identifier des gènes efficaces contre le champignon de la jambe noire Leptosphaeria maculans, nous avons transformé un colza canola (Brassica napus) avec quatre gènes de pois (Pisum sativum) sous régulation constitutive du promoteur CaMV 35S, à savoir les gènes PR10.1, chitinase, DRR206 et défensine. Les lignées transgéniques contenant des insertions d'ADN-T à copie unique pour chaque gène ont été criblés en vue de rechercher une résistance présente à la fois sur les semis (cotylédonaire) et sur la plante adulte. Les lignées transgéniques présentant une expression d'ARNm de DRR206 ou de défensine de pois ont montré des taux de maladie réduit après inoculation de L. maculans ou de Sclerotinia sclerotiorum, comparés aux lignées transgéniques n'exprimant pas ces gènes. On n'a en revanche constaté que peu voire pas d'augmentation de la résistance dans les lignées transgéniques exprimant PR10 et la chitinase. La résistance à L.maculans a en outre fait preuve d'une co-ségrégation avec les transgènes DRR206 et défensine. Des extraits de plantes transgéniques DRR206 et défensine ont produit une inhibition de la prolifération fongique in vitro. Dans les plantes transgéniques DRR206 on a également constaté une réduction de la prolifération fongique sur les sites d'inoculation, ainsi que des signes d'une réaction d'hypersensibilité.
