Definitions

  • stringent conditions is the “stringency” which occurs within a range from about Tm-5°C (5°C below the melting temperature (Tm) of the probe) to about 20°C to 25 °C below Tm.
  • Tm melting temperature
  • the stringency of hybridization may be altered in order to identify or detect identical or related polynucleotide sequences.
  • “Alterations” in the polynucleotide of SEQ ID NO: 2, as used herein, comprise any alteration in the sequence of polynucleotides encoding HPSP including deletions, insertions, and point mutations that may be detected using hybridization assays.
  • antibody refers to intact molecules as well as fragments thereof, such as Fa, F(ab') 2 , and Fv, which are capable of binding the epitopic determinant.
  • humanized antibody refers to antibody molecules in which amino acids have been replaced in the non-antigen binding regions in order to more closely resemble a human antibody, while still retaining the original binding ability.
  • nucleotide sequences which encode HPSP and its variants are preferably capable of hybridizing to the nucleotide sequence of the naturally occurring HPSP under appropriately selected conditions of stringency, it may be advantageous to produce nucleotide sequences encoding HPSP or its derivatives possessing a substantially different codon usage. Codons may be selected to increase the rate at which expression of the peptide occurs in a particular prokaryotic or eukaryotic host in accordance with the frequency with which particular codons are utilized by the host.
  • Altered nucleic acid sequences encoding HPSP which are encompassed by the invention include deletions, insertions, or substitutions of different nucleotides resulting in a polynucleotide that encodes the same or a functionally equivalent HPSP.
  • the encoded protein may also contain deletions, insertions, or substitutions of amino acid residues which produce a silent change and result in a functionally equivalent HPSP. Deliberate amino acid substitutions may be made on the basis of similarity in polarity, charge, solubility, hydrophobicity, hydrophilicity, and/or the amphipathic nature of the residues as long as the biological activity of HPSP is retained.

  • HPSP expression of HPSP is accomplished by subcloning the cDNAs into appropriate vectors and transforming the vectors into host cells.
  • the cloning vector, pSport previously used for the generation of the cDNA library is used to express HPSP in E. coli. Upstream of the cloning site, this vector contains a promoter for ⁇ -galactosidase, followed by sequence containing the amino-terminal Met, and the subsequent seven residues of ⁇ -galactosidase. Immediately following these eight residues is a bacteriophage promoter useful for transcription and a linker containing a number of unique restriction sites.
