Se han identificado dos polimorfos A y B de 1-(difenilmetil)-4-[3-(2-fenil-1,3-dioxolan-2-il)propil]piperazina. Se describe un procedimiento para la obtención de ambos y su uso posterior.

Description

POLIMORFOS A Y B DE 1- (DIFENILMETIL) -4- [3- (2-FENIL-1.3- DIOXOLA -2-IL)PROPIL]PIPERAZINA
La presente invención se refiere a nuevos polimorfos A y B de 1- (Difenilmetil) -4- [3- (2-fenil-l,3-dioxolan-2- il)propil]piperazina, compuesto que se conoce como Dotarizina (OMS) .
Breve descripción de las figuras
La Figura 1 representa el Espectro de IR del polimorfo A de Dotarizina en el rango de 4000 a 400 cm"1.
La Figura 2 representa el Espectro de IR del polimorfo A de Dotarizina en el rango de 2000 a 400 cm'1.
La Figura 3 representa el Termograma de "Differential Scanning Calorimetry" (DSC) del polimorfo A de Dotarizina. La Figura 4 representa el Difractograma de Rayos X del polimorfo A de Dotarizina.

Descripción detallada de la Invención
El compuesto 1- (Difenilmetil) -4- [3- (2-fenil-l,3- dioxolan- 2 - il ) propil ] piperazina tiene acción antiserotoninérgica (5HT1C y 5HT2) y antagonista del calcio. Es eficaz en la profilaxis de la migraña y en el trabamiento del vértigo. La obtención de este compuesto está descrita en la patente EP 97340, asignándosele en ella un punto de fusión de 93-97°C. Los solicitantes han descubierto que Dotarizina presenta dos nuevos polimorfos, A y B, cuyos puntos de fusión están ambos entre 100 y 103°C. Los termogramas de "Differential Scanning Calorimetry" (DSC) de ambos polimorfos no presentan diferencias significativas, según se ilustra en las Figuras 3 y 7. No obstante, los solicitantes han descubierto que los espectros de IR sí presentan diferencias significativas especialmente en el rango 1100 - 900 cm'1 según puede apreciarse en los espectros de IR ampliados de las Figuras 2 y 6. El polimorfo A de Dotarizina se caracteriza por presentar bandas a 2962, 2949, 2808, 2767, 1446, 1040, 1025, 1002, 993, 970, 960, 761 y 707 cm"1 y el polimorfo B se caracteriza por presentar bandas a 2961, 2883, 2812, 2777, 2760, 1448, 1283, 1058, 1042, 1029, 1006, 978, 951, 758 y 703 cm'1 (Figuras 1 y 5) . La presente solicitud presenta un procedimiento para obtener selectivamente las dos formas polimórficas de la Dotarizina. En efecto, la forma A se obtiene por cristalización en metanol, en tanto que la forma B se obtiene por cristalización en n-hexano.
Es recomendable el empleo del polimorfo A para la preparación de composiciones farmacéuticas en forma de gotas (suspensiones) , en tanto que es preferible el uso del polimorfo B para preparar cápsulas y otras formas sólidas.
Los polimorfos A y B de Dotarizina, mezclados con los excipientes farmacéuticamente aceptables, pueden administrarse a dosis diarias comprendidas entre 50 y 150 mg. Los siguientes ejemplos ilustran la obtención de los polimorfos A y B de Dotarizina y de formulaciones farmacéuticas que los contienen. Los ejemplos no se orientan a limitar la amplitud de la invención, según se ha definido, o como se reivindica más adelante.
Ejemplo 1 PolimorfoAde 1- (Difenilmetil) -4- [3- (2-fenil-l, 3-dioxolan-2- il)propil]piperazina
Se disuelven 11,2 g de Dotarizina en 56 mi de metanol a reflujo. Se filtra la disolución formada y se deja cristalizar a temperatura entre 20 y 25°C con agitación suave. Se forma un sólido cristalino que una vez seco pesa 9,9 g (rendimiento 88,5 %) . Espectro de IR (KBr) , rango de 4000 a 400 c "1: Figura 1

Ejemplo 2
PolimorfoBdel- (Difenilmetil) -4- [3- (2-fenil-1,3-dioxolan-2- il)propil]piperazina
Se disuelven 10 g de Dotarizina en 100 mi de n-hexano a reflujo. Se filtra la disolución formada y se deja cristalizar a temperatura entre 20 y 25°C con agitación. Se forma un sólido cristalino que una vez seco pesa 7,6 g
(rendimiento 76 %) .
