Abstract

La présente invention décrit un procédé de détermination du niveau d'activation des plaquettes et autres types de cellules du sang qui sont soumises à une activation par la technique ELISA ou par d'autres techniques de dosage immunologique. Ce procédé consiste à faire réagir un échantillon contenant les cellules du sang dans une phase liquide avec une quantité excessive d'anticorps primaires spécifiques à l'activation avant de permettre aux cellules dans l'échantillon de se lier à une surface solide. Cette procédure diffère des procédures traditionnelles d'ELISA, dans lesquelles les cellules sont fixées à la surface d'un tube à essai en plastique ou à une plaque de microtitrage avant d'ajouter l'anticorps primaire. Selon l'invention, il s'est révélé que le fait de lier les cellules à une surface avant la réaction avec l'anticorps primaire modifie le niveau d'activation des cellules rendant ainsi le dosage moins précis. En outre, la technique ELISA sur les plaquettes entières peut, tout comme les analyses cytométriques de flux, détecter par sensibilité les plaquettes activées.
