Abstract
Procédé permettant l'isolement et la caractérisation de gènes de C. glutamicum mis en oeuvre dans la biosynthèse d'acides aminés spécifiquement codant hom, thrB et thrC et de séquences régulant leur expression. Sont également décrites des techniques permettant de modifier ou de remplacer ces séquences ainsi qu'un moyen facilitant davantage les isolations et caractérisations, comprenant des vecteurs de sonde activateurs, utiles dans la sélection d'activateurs et de répresseurs hautement efficaces, et pouvant être régulés. On a constitué une nomenclature génomique de C. glutamicum par clivage d'ADN chromosomique avec des enzymes de restriction, par introduction de fragments d'ADN dans un vecteur approprié et par transport des molécules (rDNA) recombinantes ainsi obtenues en C. glutamicum. On a isolé des gènes hom, thrB et thrC biosynthétiques d'acides aminés codant respectivement la déshydrogénase d'homosérine, la kinase d'homosérine et la synthétase de thréonine, par moyen complémentaire d'auxotrophes de C. glutamicum. On a sous-cloné des gènes hom-thrB sur un fragment chromosomique produit par 3,6 kb Sal1 tandis que l'activité thrC avait été isolée d'un second plasmide recombinant au sein de la nomenclature génomique, et soucloné sur un fragment produit par 2.7 kb Sph1. Les places des hom-thrB et du thrC, ainsi que les séquences régulatrices ont été définies par analyses d'enzymes, moyen complémentaire d'auxotrophes E. coli définies, et cartographie de nucléase S1 et de délétion.
