Abstract
Un instrument de mesure possède une protéine à liaison sélective réversible immobilisée sur la région de la gâchette isolée d'un transistor à effet de champ (IGFET) disposé sur un capteur. Lorsque ce capteur est immergé dans une solution, la protéine lie à la gâchette un constituant choisi de la solution, produisant ainsi un effet sur un courant circulant à travers le IGFET. Une pluralité de ces agencements protéine de liaison-IGFET peuvent être ménagés sur le même capteur, comportant les mêmes protéines de liaison avec différents coefficients de liaison KD ou bien un réseau de protéines présentant une spécificité et/ou une affinité avec les ligands différente. L'analyse par un microprosseur de la réponse du IGFET à cette liaison permet, par exemple, de déterminer la concentration du constituant dans la solution. La mise en oeuvre d'une pluralité de protéines de liaison différentes permet de déterminer la concentration de différents constituants. De la même manière, l'emploi de protéines de liaison ayant des coefficients de liaison KD différents permet d'analyser le signal de sortie du capteur de façon numérique ou analogique, ou les deux combinées. Pour libérer le constituant de la protéine, on applique de la chaleur au capteur au moyen d'un assemblage à résistance intégré dans la puce dudit capteur. Dans une variante, on peut appliquer une polarisation inversée à la gâchette en vue d'accélérer cette libération.
