Claims (5)
- 検出用核酸と標的核酸との間におけるハイブリダイゼーションの検出を、二本鎖に特異的に結合して蛍光を発するインターカレーターを用いて行う方法であって、
前記インターカレーターは、負電荷を有するSYBRGreenであり、
前記ハイブリダイゼーションの場を提供し、正に帯電した固相表面が設けられた反応領域内に、所定濃度のMgCl 2 を含むHEPES緩衝液を貯留又は保持させ、前記SYBRGreenの構造を安定化させた状態とするとともに、前記反応領域に存在する一本鎖に結合した前記SYBRGreen由来の蛍光に対する二本鎖に結合した前記SYBRGreen由来の蛍光がより大きくなる状態でハイブリダイゼーションを進行させた後に、前記反応領域の洗浄を行なうことなく、ハイブリダイゼーション検出を行うことを特徴とするハイブリダイゼーション検出方法。 - 前記塩を含むHEPES緩衝液の塩濃度が、略20mMに調整されていることを特徴とする請求項1記載のハイブリダイゼーション検出方法。
- 前記塩を含むHEPES緩衝液とインターカレーターとが貯留又は保持された前記反応領域内の温度を、前記ハイブリダイゼーションに適した温度に調節する手順を含むことを特徴とする請求項1記載のハイブリダイゼーション検出方法。
- 前記塩を含むHEPES緩衝液のpHが7.0以下であることを特徴とする請求項1記載のハイブリダイゼーション検出方法。
- 前記インターカレーターを、次の(1)〜(3)のいずれかの段階で前記反応領域内に入れることを特徴とする請求項1記載のハイブリダイゼーション検出方法。
(1)前記反応領域内の前記塩を含むHEPES緩衝液へ前記標的核酸を添加する前段階。
(2)前記反応領域内の前記塩を含むHEPES緩衝液へ前記標的核酸を添加する段階。
(3)前記ハイブリダイゼーションが進行している段階。
