Abstract
本发明公开了一种饲料发酵剂及在饲料原料发酵上的应用。该饲料发酵剂由产朊假丝酵母和植物乳杆菌的发酵液等比例混合而成。将饲料原料按比例混合,原料的料水比为7:3,将1.0%葡萄糖和0.2%的K2HPO4加入水中溶解,并按5%接种量将饲料发酵剂加入水中,所得混合物加入发酵原料中,搅拌均匀装入密闭容器中室温发酵2‑3d,既得微生物发酵饲料。此方法可在室温和开放的条件下将适口性差、营养价值低的饲料原料发酵成酸香可口的微生物饲料,提高饲料营养价值和消化吸收率,调节鱼虾肠道微生物菌群。该方法操作工艺简单、成本低,利于规模推广。

技术领域
本发明涉及微生物应用和饲料发酵领域,特别涉及一种由产朊假丝酵母菌和植物乳杆菌组成的饲料发酵剂及在饲料原料发酵上的应用。
背景技术
饲料原料经微生物发酵后,原料中的蛋白质、脂肪、碳水化合物等大分子物质被微生物酶水解为氨基酸、脂肪酸、糖等小分子物质,提高了产品的营养价值和消化性能;而原料中难消化的胀气性寡糖和抗营养因子可被发酵除去或降低。因此,发酵饲料具有适口性好、营养价值高、安全、无污染等特点。饲喂发酵饲料不仅能提高饲料的消化利用率、促进养殖生物生长,还能降低排泄物中氮、磷含量,减少对环境的污染。发酵饲料中的有益微生物还能调节养殖生物肠道菌群,维持肠道微生态平衡,提高动物免疫力、减少疾病发生,降低死亡率,可减少或避免抗生素的使用,提高产品品质,利于绿色健康生态养殖的发展。
发酵饲料在畜禽养殖中的应用已经比较成熟,而在水产养殖上的应用则处于起步阶段。酵母菌和乳酸菌在水产养殖中常用作水质调节剂,也有用作拌料饲喂的研究报道,未见用于饲料或饲料原料发酵的研究报道。产朊假丝酵母和植物乳杆菌是饲料添加剂中允许添加的微生物菌种,产朊假丝酵母的菌体蛋白高、代谢产物丰富,有氨基酸、有机酸、肽等,常被用作畜禽饲料蛋白,在水产上可替代部分鱼粉。植物乳杆菌发酵过程中产生乳酸、乙酸、 丙酸和细菌素等多种物质,这些物质除具有杀毒抑菌作用外,还具有调节肠道菌群、促进消化吸收等功能,常用于牛奶等奶制品及饲料发酵当中。本发明将产朊假丝酵母和植物乳杆菌混合制备一种饲料发酵剂,并将其用于饲料原料的发酵,以提高饲料的适口性和营养价值。
发明内容
本发明公开了一种由产朊假丝酵母和植物乳杆菌及其发酵产物组成的饲料发酵剂。该饲料发酵剂由产朊假丝酵母和植物乳杆菌的发酵液等比例混合而成;所述的产朊假丝酵母保藏号:CGMCC2.281;植物杆菌保藏号:CGMCC1.511,产朊假丝酵母菌的浓度达到3.0×108-5.0×108CFU/mL,植物乳杆菌浓度达到1.0×109-5.0×109CFU/mL。
本发明进一步公开了由产朊假丝酵母和植物乳杆菌及其发酵产物组成的饲料发酵剂按以下步骤进行:
(1)产朊假丝酵母发酵:将处于对数生长期的产朊假丝酵母菌菌种按5%接种量接种到灭菌YPD液体培养中,25-28℃振荡培养20-24h。
(2)植物乳酸菌发酵:将处于对数生长期的植物乳杆菌菌种按10%接种量接种到装有90%MRS灭菌培养基的玻璃输液瓶中,30-34℃静置培养24-30h。
(3)混合发酵剂:将发酵好的产朊假丝酵母发酵液和植物乳杆菌发酵液按体积比1:1混合得到饲料发酵剂。
本发明更进一步公开了饲料发酵剂在饲料原料发酵中的应用,它是由以下技术方案实现:
(1)饲料原料有以下组合: a)豆粕、葡萄糖1.0%、K2HPO40.2%,b)花生粕、葡萄糖1.0%、K2HPO40.2%,c)豆粕20%、麸皮80%、葡萄糖1.0%、K2HPO40.2%,d)豆粕20%、豆皮80%、葡萄糖1.0%、K2HPO40.2%,e)全价料、葡萄糖1.0%、K2HPO40.2%。
(2)上述原料的料水比为7:3,接菌量5%,接种混匀后置于密闭容器中室温(20-30℃)发酵,发酵2d后取样,测pH、乳酸菌、酵母菌含量。
(3)将发酵好的物料即为微生物发酵饲料,可直接投喂,也可风干装入袋中。
本发明更进一步公开了采用饲料发酵剂制备的微生物发酵饲料在调节鱼虾肠道微生物菌群方面的应用。特别是在降低细菌总数、弧菌总数和弧菌有害菌的比例,增加有益菌的比例方面的应用,实验结果显示:处理组的细菌总数和弧菌总数低于对照组,弧菌仅占总细菌数的12.3%,而对照组的弧菌总数占24.0%。
本发明曾经考察过芽胞杆菌、乳酸菌、酵母菌三种菌混合作为饲料发酵剂进行发酵,由于饲料原料发酵后pH值降低,芽胞杆菌基本不生长;也考察了以芽胞杆菌、乳酸菌、酵母菌的单一菌种作为发酵剂进行发酵,饲料发酵后的气味、色泽和有效活菌浓度等指标,不如乳酸菌和酵母菌混合发酵。上述试验主要解决了混合发酵剂的组成问题。
本发明公开的饲料发酵剂及在饲料发酵上的应用与现有技术相比,所具有的积极效果在于:
(1)本发明采用产朊假丝酵母、植物乳杆菌、葡萄糖、磷酸氢二钾与豆粕、麸皮、花生粕、豆皮、全价料混合,在室温和开放的条件下即可完成发酵,操作工艺简单、成本低,利于规模推广。
(2)本发明提供的菌种在生长、产酸、抑菌等方面表现良好的性能,发酵时间短、所需温度低,即室温(20-30℃)2d即可完成发酵,发酵后的饲料pH降低到4.02以下,发酵后的饲料有效活菌浓度高,在不风干的情况下室温放置30d无杂菌生长。
(3)本发明采用简单工艺即可使产朊假丝酵母和植物乳杆菌在饲料原料中迅速增殖,并使适口性差、营养价值低的饲料原料发酵成酸香可口的微生物饲料,具有诱食效果并能提高饲料营养价值和消化吸收率,调节鱼虾肠道微生物菌群。

下面结合具体实施例对本发明进行详细说明。以下的实施例将有助于本领域的技术人员进一步理解本发明,但不限制本领域的专业人员按照本发明的精神对其进行改进和变化,所述的这些改进和变化都应视为在本发明的范围内,本发明的范围和实质由权利要求来限定。所述的产朊假丝酵母保藏号:CGMCC2.281;植物杆菌保藏号:CGMCC1.511。
所述的YPD培养基用来培养酵母菌,主要成分及含量为酵母膏10g/L,葡萄糖20g /L,蛋白胨20g/L;MRS培养基用来培养乳酸菌,主要成分及含量为蛋白胨10g/L,牛肉膏10g/L,酵母膏5g/L,葡萄糖20g/L,柠檬酸氢二铵2g/L,乙酸钠5g/L,磷酸氢二钾2g/L,硫酸镁0.58g/L,硫酸锰0.25g/L,吐温80 1mL/L。
实施例1
一种饲料发酵剂,其特征在于该饲料发酵剂由产朊假丝酵母和植物乳杆菌的发酵液等比例混合而成;所述的产朊假丝酵母保藏号:CGMCC2.281;植物杆菌保藏号:CGMCC1.511,产朊假丝酵母菌的浓度达到3.0×108-5.0×108CFU/mL,植物乳杆菌浓度达到1.0×109-5.0×109CFU/mL;其制备步骤如下:
(1)产朊假丝酵母发酵液制备
将斜面保存的酵母菌菌种,接种于新鲜的YPD培养基平板上于25℃培养48h。从平板上挑取一环,接种于装有100mL种子培养基的250mL的锥型瓶中,25℃,150r/min摇床,振荡培养24h。
(2)植物乳杆菌发酵液制备
将穿刺培养的乳酸菌菌种,接种于新鲜的乳酸菌培养基平板上于30℃厌氧培养72h。从平板上挑取一环,接种于装有45mL种子培养基的50mL的厌氧瓶中,30℃,静置培养48h,再将其以10%的接种量接种于装500mL培养基的输液瓶中,30℃静置培养24h。
(3)饲料发酵剂的制备
将产朊假丝酵母菌发酵液和植物乳杆菌发酵液以1:1混合,既得饲料发酵剂。
按上述方法制得的饲料发酵剂,产朊假丝酵母菌的浓度达到3.8×108CFU/mL以上,植物乳杆菌浓度达到2.5×109CFU/mL以上。
实施例2
饲料发酵剂在饲料原料发酵中的应用,操作步骤如下:
(1)饲料原料有以下组合:a)豆粕、葡萄糖1.0%、K2HPO40.2%,b)花生粕、葡萄糖1.0%、K2HPO40.2%,c)豆粕20%、麸皮80%、葡萄糖1.0%、K2HPO40.2%,d)豆粕20%、豆皮80%、葡萄糖1.0%、K2HPO40.2%,e)全价料、葡萄糖1.0%、K2HPO40.2%。
(2)上述原料的料水比为7:3,将葡萄糖和K2HPO4加入水中溶解,并按5%接种量将饲料发酵剂加入水中,得混合物A,将A加入发酵原料中,搅拌均匀的混合物B。
(3)将B装入密闭性好的塑料圆桶或塑料袋中,室温(20-30℃)发酵,饲料原料散发明显的酸香味完成发酵,一般只需2d,发酵后原料中乳酸菌浓度达到6.74×109CFU/g湿重以上,酵母菌浓度达到1.45×108CFU/g湿重以上,pH降到4.02以下(表1)。

(4)发酵好的饲料可按比例加入到常规饲料中直接投喂,也可风干后装入袋中日后饲喂。
实施例3
饲料原料发酵后使用情况
方法:试验挑选外观正常、体质健壮的南美白对虾作为实验用虾,每个池(20m2)放养南美白对虾5000尾,每天按体重 6%投饵,分四次投喂,对照组投喂常规饲料,处理组将发酵好的饲料按10%比例添加到常规饲料中饲喂。饲养期水温(29±3)℃,连续充气。试验第6周,取肠道,去除内含物,用去离子水冲洗后滤纸吸干,称重,捣碎,加入10mL无菌生理盐水中匀浆,梯度稀释后测细菌总数和弧菌数。
结果:从表2可知,添加发酵饲料的处理组,其细菌总数和弧菌总数低于对照组,弧菌仅占总细菌数的12.3%,而对照组的弧菌总数占24.0%。说明添加微生物发酵饲料能改善肠道菌群,降低有害菌的比例,从而增加有益菌的比例。

注:菌落总数以每克肠道的相应微生物数量的对数表示,即lg( cfu/ g)。
Claims (6)
1.一种饲料发酵剂,其特征在于该饲料发酵剂由产朊假丝酵母和植物乳杆菌的发酵液等比例混合而成;所述的产朊假丝酵母保藏号:CGMCC2.281;植物杆菌保藏号:CGMCC1.511,产朊假丝酵母菌的浓度达到3.0×108-5.0×108CFU/mL,植物乳杆菌浓度达到1.0×109-5.0×109CFU/mL。
2.权利要求1所述饲料发酵剂的制备方法,其特征在于按如下的步骤进行:
(1)产朊假丝酵母发酵:将处于对数生长期的产朊假丝酵母菌种按5%接种量接种到灭菌YPD液体培养中,25-28℃振荡培养20-24h;
(2)植物乳酸菌发酵:将处于对数生长期的植物乳杆菌菌种按10%接种量接种到装有90%MRS灭菌培养基的玻璃输液瓶中,30-34℃静置培养24-30h;
(3)混合发酵剂:将发酵好的产朊假丝酵母发酵液和植物乳杆菌发酵液按体积比1:1混合,制得饲料发酵剂。
3.一种采用权利要求1所述的饲料发酵剂对饲料原料发酵的方法,其特征在于按如下的步骤进行:
(1)将饲料原料按比例混合,原料的料水比为7:3;
(2)将1.0%(w/w)葡萄糖和0.2%(w/w)的K2HPO4加入水中溶解,并按5%(w/w)接种量将饲料发酵剂加入水中,制得混合物A;
(3)将所得混合物A加入到步骤(1)的待发酵原料中,搅拌均匀装入密闭容器中室温发酵2-3d,测pH、乳酸菌、酵母菌含量,得到微生物发酵饲料;发酵后原料中乳酸菌浓度达到6.74×109CFU/g湿重以上,酵母菌浓度达到1.45×108CFU/g湿重以上,pH降到4.02以下。
4.权利要求1所述的饲料发酵剂在提高饲料原料发酵效率方面的应用。
5.采用权利要求3所述方法制备的微生物发酵饲料在调节鱼虾肠道微生物菌群方面的应用。
6.权利要求5所述的应用,其中调节鱼虾肠道微生物菌群指的是:降低细菌总数、弧菌总数和弧菌有害菌的比例,增加有益菌的比例。
