The adhesion receptor CD2 plays an important role in the full activation of T cells. Dustin and colleagues show that CD2 occupies a region in the periphery of the immunological synapse where it amplifies cognate antigen signals, whereas the presence of PD-1 disrupts this effect.

T_FH-derived dopamine accelerates productive synapses in germinal centres

Human follicular helper T cells engaging in synaptic interactions with germinal centre B cells release dopamine stored in chromogranin B⁺ granules, causing rapid externalization of ICOS ligand, which in turn enhances CD40L delivery to the synaptic cleft and synaptic contact, and results in an accelerated response.

Spatial dysregulation of T follicular helper cells impairs vaccine responses in aging

Linterman and colleagues examine germinal center formation in older individuals. They find that aged T_FH cells have dysregulated CXCR4 expression, which causes spatial mislocalization of these cells in germinal centers, impairing their ability to provide help to B cells and to promote antibody production.

High SARS-CoV-2 seroprevalence in children and adults in the Austrian ski resort of Ischgl

Knabl et al. perform a seroepidemiological study in the Austrian ski resort Ischgl, where a super-spreader event lead to a SARS-CoV-2 outbreak. Through mathematical modelling, they find that the subsequent decline in viral transmission was most likely a combined effect of high seropositivity and the implementation of non-pharmaceutical interventions.

Improving personalized tumor growth predictions using a Bayesian combination of mechanistic modeling and machine learning

Mascheroni et al. develop a method for individual clinical predictions by combining mathematical modelling and machine learning in a Bayesian framework (BaM³). By using both synthetic and real clinical datasets, they show the potential of the method to predict tumour growth in the context of clinical data sparsity and limited knowledge of disease mechanisms.
